The agar solution doesn't necessarily need to be boiled to dissolve (as a first step); as long as it is thoroughly mixed, the autoclave's heat and boiling of the water during sterilization will cause it to go into solution. Tygar 07:13, Feb 12, 2005 (UTC)

Suggested move to wikibooks

I disagree that this article should be moved to wikibooks as per the recently added tag. It may well be that the article can be improved and made more encyclopaedic and that some of the "how to" might belong elsewhere, but as a frequent user of wikipedia, I was searching for this infomation here and expected to find it on wikipedia--User:AYArktos | Talk 01:42, 20 September 2005 (UTC)

Question about incubation times

How long are the plates usually incubated for? I assume it depends on the experiment. --User:AYArktos | Talk 01:50, 20 September 2005 (UTC)

From distant memory, when looking for most human pathogens, plates are incubated for two days in total, although many are looked at after one day, before being reincubated.

For slower growing pathogens (e.g Campylobacter, Bordetella), plates are sometimes incubated for longer. But for commoner Staphs, Streps, candida, respiratory bugs, gram negative bacilli, etc., two days is sufficient. Every lab will have their own protocols however. Streaking

At the top of the page, only streaking is mentioned. What about spread plates and pour plates? This is an agar plate page, and I think the streaking references do not belong, or if they do, should be complemented in a separate section by the other methods I mentioned. Incorrect use of the plural

"A... media" appears several times throughout this article (e.g. "A selective and differential media..."). "Media" is the plural of "medium". Each instance should be corrected to "a... medium". 71.96.70.9 (talk) 20:17, 8 February 2009 (UTC)

Not an agar plate after microorganisms have been added?

"An agar plate is a sterile Petri dish that contains a growth medium (typically agar plus nutrients) used to culture microorganisms or small plants like the moss Physcomitrella patens."

Casein nutrient agar

Having edited the See also section, to include a link to the previously orphaned Casein nutrient agar article, and grouped it with two other entries for recipes of agar that had already been in the list, I now see there is a template for { { Growth media } } (which, of course does not list Casein nutrient agar, hence it previously having been an orphan article). I think I had better leave all this to an editor more expert than I to sort out :-) TheAMmollusc (talk) 07:38, 30 January 2013 (UTC)
