By inducing G ([[guanine]]) to T ([[thymidine]]) [[transversion]]s in transversion hotspots within [[p53]], there is a probability that benzo[''a'']pyrene diol epoxide inactivates the tumor suppression ability in certain cells, leading to cancer.

As illustrated above, (+)benzo[a]pyrene-7,8-dihydrodiol-9,10-epoxide (BPDE) forms bulky covalent [[DNA adduct]]s with [[guanine]]s.
